Although not strictly a "hitman", I'd nominate Tommy from Goodfellas. Ok, he's more of a henchman but he does hits. And possibly the only person to kill Samuel L Jackson (apart from the Mensa Shark in Deep Blue) on film.

No 1 is the right choice. I'd find a place for Havey Keitel's cleaner in The Assasin, pouring acid down someones throat is nastier than anything on this list.
Oh and that Eric Bana quote is from Knocked Up. Tut tut
